Actividad antioxidante de flavonoides del tallo de orégano mexicano (Lippia graveolens HBK var. berlandieri Schauer)

México es el principal exportador a nivel mundial de orégano mexicano (Lippia graveolens), con 35 – 40 % del mercado internacional; su alta demanda se debe al contenido de aceite esencial de calidad en la hoja. Su recolección es complementaria a la agricultura de temporal o secano en zonas áridas y semiáridas. El desaprovechamiento del tallo y los escasos estudios fitoquímicos y farmacológicos reportados, indujeron esta investigación en la que determinó el tipo de flavonoides y se evaluó la actividad antioxidante de extractos crudos del tallo de Lippia graveolens H.B.K. var. berlandieri Schauer recolectado en el Mezquital, Durango, México. Los extractos se prepararon por maceración del tallo molido a temperatura ambiente con extracciones sucesivas de hexano, acetato de etilo y metanol. El extracto acuoso se preparó por ebullición del tallo en agua (10 %) con posterior liofilización. Los extractos fluidos de acetato de etilo y de metanol se fraccionaron en columnas de cromatografía convencional sobre silicagel-G. Dos fracciones del extracto de acetato de etilo y una del extracto de metanol se analizaron por cromatografía de líquidos acoplada a espectrometría de masas. Se detectó pilosina, cirsimaritina, naringenina, kaempferol, isokaempferido, un derivado de catequina y un hexósido de quercetina no identificado. La actividad antioxidante se evaluó en los extractos por el método del radical libre 2,2-difenil-1-picrilhidracilo (DPPH). La reducción del radical con los extractos de prueba fue mayor a 91.2 % en 100 mgL-1 excepto para el extracto acuoso que fue de 87.6 % en 50 mgL-1, Los valores de IC50 fueron relevantes para el extracto de acetato de etilo (sólido: 11.24 ± 1.10; extracto fluído: 12.94 ± 0.63). Estos resultados muestran la presencia en el tallo de moléculas bioactivas útiles para el desarrollo de fármacos derivados de productos naturales.

The aqueous extract was prepared by boiling the stem in water (10 %) and then liofilized. The fluid extracts of ethyl acetate and methanol were fractionated by column chromatography on silica gel G. Two fractions of the ethyl acetate extract and one of the methanolic extract were analized by liquid chromatography coupled to mass spectrometry. We detected pilosin, cirsimartin, narigenin, kaempferol, isokaemferide, a derivative of catechin and a non identified. hexoside of quercetin The antioxidant activity of the extracts was evaluated by the method of the radical 2,2-diphenyl-1-picryl hydracyl (DPPH).
