Effects of purslane herb on stress ability of aging mice induced by D-galactose

Objective: To investigate the effects of purslane herb aquenous extracts (PHAS) on the stress ability of aging mice induced by D-galactose.

Methods: We odserved the survival time to hypoxia and heat survival rate of the mice treated with different doses of PHAS and vitamin E. The contents of lipofuscin and malondialdehyde (MDA), and the activity of superoxide dismutase (SOD) and catalase (CAT) in the brain and liver of the mice were tested.

Results: As compared with vitamin E, three doses of PHAS (1.6, 0.8 and 0.4 ml/d) prolonged the survival time to hypoxia and the pole climbing time and increased the heat survival rate, and the 0.8 ml/d PHAS had the best effect.In the group of 0.8 ml/d PHAS, the activity of SOD and CAT decreased less, and the contents of lipofuscin and MDA decreased significantly.The effect of vitamin E was not as good as the PHAS.

Conclusion: PHAS can prevent the stress ability of the aging mice.One of its mechanisms may be increasing the activity of SOD and CAT, hence decreasing the damage of the oxidation products to the body.

     大量研究表明,老年机体对外环境刺激的应激能力减弱 [1],提高老年机体的应激能力是抗衰老研究领域的热点问题。马齿苋是一味清热解毒的中药,也是一种深受欢迎的野菜,《本草纲目》称之为长命菜 [2]。近年来有人发现马齿苋含有丰富的α-亚麻酸和维生素E、胡萝卜素等,并且对老年人常见的高血脂、高血糖等病症有防治作用 [3,4]。本实验对马齿苋是否能够提高老年机体的应激能力进行了初步探索,现将研究结果报道如下。

 
   
1  材料和方法
1.1  药物
  马齿苋全草 (干)购自上海市药材公司,按文献方法水提醇沉 [5],制成浓度为含生药1 g/ml的水提液备用。维生素E粉剂为北京天天维他保健食品有限公司产品,纯度为50%,加蒸馏水配制成浓度为0.6 mg/ml水溶液备用。
1.2  实验动物分组和衰老模型制作  清洁级10周龄BALA/C雄性小鼠, 体重(21±2) g,购自上海西普尔-必凯实验动物有限公司(沪动合证字153号)。按体重随机分为正常组、衰老模型组、衰老模型+维生素E组和衰老模型+马齿苋组。除正常组以外,其余各组小鼠参照有关文献 [1],按体重腹腔注射D-半乳糖(Sigma公司产品,第二军医大学附属长海医院制剂室制成10 mg/ml针剂)0.1 mg·g-1·d-1,连续40 d,当小鼠出现脊椎隆起,体形消瘦,毛色灰暗稀疏,行动迟缓等老年体征,证明造模成功。在造模的同时,模型组灌喂蒸馏水0.8 ml/d,维生素E组灌喂浓度为0.6 mg/ml 维生素E溶液0.8 ml/d,马齿苋组分别灌喂浓度为1g/ml的马齿苋水提液1.6、0.8、0.4 ml/d,均连续灌喂40 d。
1.3  观察指标和方法
1.3.1  抗疲劳能力
  于末次灌胃30 min后,将各组小鼠置于直径0.8 cm,长度60 cm,上端固定,下端悬空的光滑玻璃棒上,下面放置水盆(水温15 ℃,水深10 cm),记录小鼠从上玻璃棒到滑落的时间,重复3次,取平均值为爬杆时间(判断肌疲劳)。
1.3.2  高温存活率  末次灌喂30 min后,将各组小鼠放入42 ℃、相对湿度80%的恒温箱热暴露1 h,计算小鼠存活率。
1.3.3  缺氧生存时间  于末次灌胃30 min后,将各组小鼠放入150 ml广口瓶中(内置10 g钠石灰和滤纸),每瓶1鼠,瓶塞用凡士林密封,观察小鼠从瓶塞密封到呼吸停止的时间,作为缺氧生存时间。
1.3.4  抗氧化指标  采用荧光法测定脑和肝脏脂褐质、丙二醛(malondialdehyde, MDA)含量;采用羟胺法测定脑、肝超氧化物歧化酶(superoxide dismutase, SOD)活性;采用钼酸胺显色法测定过氧化氢酶(catalase, CAT)活性,上述实验所用试剂盒均购自南京建成生物工程研究所。
1.4  统计学方法  采用SPSS 11.0软件进行F 检验。

2  结 果
2.1  马齿苋对D-半乳糖衰老模型小鼠应激能力的影响
 模型组小鼠的爬杆时间、缺氧生存时间和高温存活率均比正常组小鼠明显减少。灌喂马齿苋的各组小鼠与正常组小鼠相比,爬杆时间和缺氧生存时间没有明显变化,高温存活率虽比正常组减少,但减少幅度比模型组明显减小,尤其是中剂量和高剂量组的存活率仅减少了20%。而维生素E组小鼠的爬杆时间、缺氧生存时间和高温存活率与模型组比较没有明显差别。见表1。

2.2  马齿苋对D-半乳糖衰老模型小鼠抗氧化能力的影响  见表2、表3。衰老模型组小鼠大脑皮层和肝细胞的SOD,及肝细胞和红细胞CAT活性比正常组明显降低,而大脑皮层和肝细胞的MDA、脂褐素含量却比正常组明显升高。马齿苋高、中剂量组小鼠大脑皮层和肝脏细胞的SOD活性、肝细胞和红细胞CAT活性比模型组明显提高,而MDA、脂褐素含量与模型组比较明显降低,但马齿苋低剂量组SOD和CAT活性,以及MDA、脂褐素含量与模型组比变化不大。维生素E对衰老模型小鼠SOD和CAT活性虽也有一定的保护作用,但作用强度不如高、中剂量马齿苋。维生素E使衰老模型小鼠肝细胞中的MDA含量有明显下降。

3  讨 论
     马齿苋是一味常用中药,具有清热解毒,消炎止痛的作用。近年来人们发现马齿苋有抗氧化和消除自由基的作用 [4]。我们的实验发现,给D-半乳糖衰老模型小鼠长期(40 d)灌喂马齿苋水提液可以明显延长其爬杆时间和缺氧生存时间,提高其高温存活率。
     人体的抗疲劳和抵抗恶劣环境等生理功能都会随着年龄的增长而逐渐下降。有研究表明机体清除自由基的能力也随着年龄增长而下降 [6,7],特别是 SOD和CAT的活性可明显下降,导致体内自由基产生增多,而过多的自由基会损伤细胞膜,破坏蛋白质、脂肪和核酸,形成脂褐质和MDA等物质,影响细胞的寿命,最终引起机体衰老。我们的实验结果也发现,注射40 d D-半乳糖后小鼠大脑、肝脏和红细胞的SOD和CAT活性明显降低,而脂褐质和MDA的含量明显升高。但有意义的是,给注射D-半乳糖的小鼠同时灌喂马齿苋水提液,可明显减小SOD和CAT活性下降的幅度,进而减少脂褐质和MDA在细胞内的堆积。这些现象提示,马齿苋可能通过减轻D-半乳糖对小鼠大脑、肝脏和红细胞的SOD和CAT活性的损伤,保护了机体清除自由基的能力,从而减慢了衰老的进程。当然,衰老是一个极其复杂的综合过程,尤其是人类的衰老更是涉及到机体各个组织器官的生理病理变化。另外,马齿苋的成分也比较复杂,除了有抗氧化物质以外,还有其他许多生物活性物质。因此,野生植物马齿苋对人类衰老过程的干预作用以及具体的作用机制还有待深入研究。
