Differentiation of biologically distinct Cucumber mosaic isolates by double stranded RNA profile
Abstract
Four isolates of Cucumber mosaic virus from Nicotiana tabacum, Commellina species and Petunia, designated as MV- T, C, PI and P 2 , differed in their host reaction, temperature sensitivity, and at the molecular level. Serologically related isolates of CMV viz. CMV-P I , P 2 , C and T were analyzed on the basis of their symptomology, temperature tolerance and double stranded RNA pattern. Double stranded RNA (ds-RNA) pattern of these CMV isolates representing biological diversity were examined. Two distinct patterns differing by the number and or/position of the ds- RNA bands were obtained for the four isolates. The ds-RNA pattern can not be necessarily co-related with the symptoms severity nor with any specific biological characteristics, however, it can be utilized in distinguishing isolates.
